[ピアス]サーモフィッシャーサイエンティフィック Seize Primary：免疫沈降キット

抗体の種、クラス、サブクラス、またプロテインAやGとの結合価も心配無用です
AminoLinkゲルはタンパク質、ペプチド、抗体、抗原などのリガンドを、リジンやN末端の一級アミンを介して固定化する、アルデヒド基で活性化された4%架橋化アガロースビーズです。固定化はシッフ塩基形成により共有結合的に行われるためSDS-PAGEでの抗体由来の重鎖/軽鎖のコンタミもなく、調製された抗体カラムは10回程度繰り返し使用することが出来ます。

免疫沈降（IP）、免疫共沈（Co-IP）、小スケールでの精製用カラム
精製済みのアミン化合物を、架橋化剤を使用せず、簡単に共有結合的に固定
プロテインAやGに結合価の低い抗体の固定化（チキンIgY, ヒトIgE, マウスIgG2a
結合バッファーはpH7.2またはpH10から選択

Note:固定するタンパク質は精製されていることが必要です。AminoLinkゲルはアミンを含む分子に対して結合するため、トリスのようなアミン含有バッファー中のタンパク質や、ゼラチンやキャリアタンパク質を含む抗体をゲルに添加すると、これらの共存するアミン化合物と競合し、固定化反応は阻害されます。透析などによりバッファーを交換するか、精製してから添加して下さい。

Fig 1　AminoLink Plusへの哺乳類および鳥類の抗体結合効率 各種の精製抗体(200 μg)を200μlのAminoLink Plus Coupling Gelと室温で4時間(1時間毎)反応させた。（チキンIgYは500μgが使用された。）約80-88% の結合効率が4時間で得られた。

Fig 2　Seize Primary Kit (AminoLink Plus担体) によるGFPの免疫沈降 アフィニティ精製されたヤギ抗GFP抗体(200 μl)を200 μl AminoLink Plusゲルと室温で4時間反応させ87％(174 μg)の抗体がカップリングされた。GFPを過剰発現させたBL21株のB-PER Bacterial Protein Extraction Reagent (Cat#78248)により可溶化し、75 μlライセートで免疫沈降を行った。洗浄画分の5%希釈液、ライセート10%希釈液、溶出画分を4-20% PAGEゲルで電気泳動後、GelCode Blue Stain ReagentによりCBB染色に供した。マーカー(M) は TriChromRanger Prestained Protein Molecular Weight Marker Mix (Cat#26691)を使用した。
